It is absolutely necessary to confirm any positive screening result for phenylketonuria by 2 or more independent procedures carried out on 2 or more different [infant] blood specimens before initiation of diet treatment. Details for a combined paper chromatography and inhibition assay of cut paper strips for positive identification of blood phenylalanine are given. This procedure correlates the partition between 2 solvents, the reaction to ninhydrin and the reaction in inhibition assay.
