Linkage dis equilibria between the hal malignant hyper thermia phospho hexose isomerase and 6 phospho gluconate dehydrogenase loci in 2 pietrain lines

Pietrain pigs [250] from 2 lines selected for 3 generations, one (A line) on a combination of average daily gain and backfat thickness and the other (B line) on muscular hypertrophy, were submitted to a halothane-test and their genotypes determined for 2 red cell enzymes (phosphohexose isomerase [PHI] and 6-phosphogluconate dehydrogenase (6-PGD)). The occurrence in the halothane sensitive group of 11 animals heterozygous at the PHI locus definitely proves that the structural gene of this enzyme and the gene controlling MHS are located at 2 distinct loci. The 2 lines differ with respect to the linkage disequilibrium between Hal and PHI, which is very highly significant in the A line and not significant in the B line. The frequency of the Hals allele responsible for MHS is significantly higher in the B line than in the A line, assuming a penetrance of Hals equal to 0.75, which is in favor of the hypothesis of an association between muscular hypertrophy and malignant hyperthermia syndrome. The higher frequency of the PHIB allele observed in the B line compared to the A line is probably due to the linkage disequilibrium found between the Hal and PHI loci and is probably not to a real effect of the PHI locus on muscular development.
