The ability of the organophosphorus insecticide malathion (O,O-dimethyl-S-(1,2-dicarbethoxyethyl)dithiophosphate) (CAS number 121-75-5) to increase the frequency of micronuclei in mice was reported. Taking into account that micronuclei arise mainly from the induction of chromosomal or chromatid breaks, this finding could be considered as a good indication of the clastogenic capacity of the compound. Studies developed with cultured mammalian cells showed that malathion induces sister-chromatid exchanges and cell-cycle delay (Nicholas et al., 1979; Chen et al., 1981) and increases the frequency of chromatin bridges in anaphase-telophase (Dulout et al., 1978). Van Bao et al. (1974) found an increment of chromosomal aberrations in human beings acutely intoxicated with the compound. Contrary evidence was reported by Huang (1973), who claimed that the pesticide inhibited the proliferation of human lymphoid cell lines but did not increase the frequency of chromosomal aberrations. To pursue studies about the genotoxic effects of malathion, an experiment based on the analysis of bone-marrow metaphase chromosomes of mice treated with low doses of the compound and killed at various times after treatment was developed.
