On the amino acid composition in soil hydrolysates part 2 amino acid composition in acid hydrolysates of soil organic matter fractionated according to its size and density

Wet soil samples (F- and H-horizon) of podzolic soil were fractionated by sieving, weighing density and centrifugal sedimentation, successively; for the F-horizon, 6 fractions, and for the H-horizon, 5 fractions were obtained. The amino acid compositions in each fraction were estimated by their acid hydrolysates. The C/N ratio of each fraction in F-horizon and H-horizon decreased with diminishing size and the C/N ratio of the F-horizon was higher than that of the H-horizon. The .alpha.-amino N content of each fraction in the F-horizon ranged from 50.1-29.5% to total N; and in the H-horizon, 47.2-21.0%. The percentage of this form of N in the F-horizon was higher than in the H-horizon and there was no apparent relationship between .alpha.-amino N content and each fraction. Sixteen amino acids and ammonia were found and 4 unidentified ninhydrin-positive compounds were detected in all fractions. The total amino acid compounds were greater in the F-horizon than in the H-horizon. Mole percent of total amount of major amino acids in the F-horizon was 57.0% and in the H-horizon, 61.7%. Total basic amino acid of each fraction in the F-horizon was higher than that in the H-horizon. The ratios of glycine to glutamic acid varied more in the F-horizon than in the H-horizon.
