Pantethine's inhibition of lipogenesis from liver pyruvate in db db mice

A study was made with C57Bl/KsJ mice in order to determine a possible mechanism of pantethine's effect on lipogenesis from liver pyruvate. The repeated administration of pantethine to hyperlipogenetic db-db mice was found to inhibit fat biosynthesis of fatty acids and lipids from liver pyruvate. It is apparently related to the inhibition of the activity of NADP-dependent cytoplasmic dehydrogenases and transhydrogenase, and to the activation of glutathione reductase. The mechanisms responsible for fatty acid and lipid biosynthesis inhibition also include an acute decrease in the levels of hepatic NADP-H, a decrease in the blood plasma corticosterone concentration, and changes in the content and structure of coenzyme A hepatocytes, related to it.
