Phage induced mutations in xanthomonas campestris pathovar citri

All the survivors of X. campestris pv. citri XW47 became lysogenic after infected with filamentous phase Cf16, among which 10% of the lysogens were Cys- mutants, and the others were prototrophs. The Cys- mutation also accompanied with changes in citrate utilization, tolerance to 3% NaCl and decomposition of galactose, trehalose, xylose, mannitol, mannose, glycerol, dextrin and raffinose. The Cys- mutants did not revert spontaneously or by treatments with acridine orange, high temperature, UV light and soft agar stab preservation, respectively, but reverted within 4 wk at the frequencies of 4.8 .times. 10-5-10-3 in average after inoculated into leaves of Liucheng sweet orange. All the revertants were cured of Cf16 prophage. Some of the revertants behaved unlike their wild-type strain in tolerance to 3% NaCl, which suggest that genetic rearrangements had taken place in these revertants after curing of Cf16. The Cys- mutants exhibited decrease in virulence, but their revertants showed the virulence same as the wild-type strain. The prototrophic lysogen showed the same reactions as its wild-type strain in most of biochemical characteristics tested and virulence in Liucheng sweet orange, except in the tolerance to 3% NaCl. The cured clones from the prototrophic lysogen, however, also changed some of biochemical characteristics.
