Plasma cyclic nucleotide levels in patients with homozygous beta thalassemia

[In agreement with the hypothesis that cAMP and cGMP play a role in the control of cell growth and differentiation; alterations in cyclic nucleotide metabolism have often been found in neoplastic tissues.] To investigate the possibility that a proliferative non-neoplastic process influences extracellular cyclic nucleotide concentrations, plasma cAMP and cGMP levels were measured in 38 patients with homozygous .beta.-thalassemia. This group consisted of 20 patients with thalassemia major transfused regularly (mean pretransfusion Hb levels, 11 g/dl) and 18 patients with thalassemia intermedia who did not require regular blood transfusion (mean Hb levels, 8.7 g/dl). In the patient group, plasma cAMP levels were similar to those of 37 normal subjects matched for age and sex, whereas plasma cGMP levels were markedly higher. Moreover, in the thalassemic patients there was a significant negative correlation between plasma cGMP levels and Hb concentrations, suggesting that their marked erythroid hyperplasia may play a role in determining alterations in extracellular cGMP levels.
