Plastein synthesis by alpha chymotrypsin immobilized on hydrophobic agarose gel

.alpha.-Chymotrypsin was immobilized on hydrophobic agarose gels (octyl-Sepharose, phenyl-Sepharose). Native and immobilized .alpha.-chymotrypsin were employed for the plastein reaction. The yield of plastein from the immobilized .alpha.-chymotrypsin was higher than that using native .alpha.-chymotrypsin. The optimum pH value of native and immobilized .alpha.-chymotrypsin for plastein synthesis was 5.5. The immobilized .alpha.-chymotrypsin was stable under alkaline conditions. The optimum substrate concentration for the immobilized .alpha.-chymotrypsin was 25%. The immobilized .alpha.-chymotrypsin can be used repeatedly for plastein synthesis. The loss of catalytic activity was within 30% after 5 use cycles. The MW of the plastein synthesized was determined by the SDS[sodium dodecyl sulfate]-polyacrylamide disc-electrophoresis method. The MW of the plastein was from 13,000-30,000.
