The 5HT-induced shape change of blood platelets, suggested to represent the 5HT receptor sensitivity, and the platelet and plasma 5-HT concentration were measured simultaneously in reserpinized rabbits. Reserpine was given i.m. acutely (one shot) or chronically (once a day for 2 weeks). The platelet 5HT concentration decreased after both acute and chronic administration of reserpine. The plasma 5HT concentration increased after the acute administration, but it remained unchanged after the chronic administration. Similarly, the 5HT-induced shape change decreased after the acute administration, but remained unchanged in the chronic administration. These results suggest that the 5HT-induced platelet shape change may depend on the 5HT concentration in the plasma but not in the platelet. Besides, these results support Pletscher's suggestion that the platelet may be considered as a model for some 5HT receptors of the central nervous system.
