Platelet size analysis platelet diameter evaluation on blood films

Evaluation of platelet diameter on blood films which were made by several technical methods was discussed. Microscopic measurements of blood films fixed with glutaraldehyde showed that in healthy subjects circulating platelets approximated ellipsoids of rotation whose diameter exceeded thickness about 4-fold and whose diameter increased about 1.5-fold after sphering by excess EDTA. The shape of circulating platelets in Wiskott-Aldrich syndrome, a platelet function disorder, was not discoid. In the typical unfixed dried EDTA blood films the platelet diameter medians were reduced by platelet shrinkage. It is extremely difficult to determine unfixed blood film diameter based on these methods or to discuss platelet size because many factors influence the platelet diameter on unfixed blood films.
