Preparation of insolubilized dna

A preliminary experiment showed that the insolubilized DNA prepared by the direct addition of multivalent metal ion to DNA solution differed greatly from that prepared by electrolysis in the amount of water absorbed and protamine combined to insolubilized DNA. To elucidate these molecular differences the conditions for insolubilization of DNA by electrolysis were investigated. The yield of insolubilized DNA was positively correlated with the increase in both concentration and pH of DNA solution, while it decreased gradually with increasing temperature. The amount of insolubilized DNA increased with increasing current passed. Cu or Ni were the preferred electrode plates. The current efficiency was dependent on the concentration of DNA solution. The formation of insolubilized DNA was not related to the current density.
