Preparation of purified a polypeptide chain from a ricin lectin
A method for the preparation of purified A-poly-peptide chain isolated from RCA2 lectin of castor bean seeds [Ricinus communis] is presented. The A-chain was homogeneous, lacking galactose-reacting active site and being devoid of any significant amount of carbohydrates. The ability of A-chain to kill in vitro EL-4 leukemia cells and its toxicity for mice were both at least 1000 times lower than those of RCA2. The A-chain regained the initial toxicity of RCA2 by covalent binding to a carrier able to attach to the cell surface via IgG antibody (e.g., protein A of Staphylococcus aureus).
