A simple and reproducible method for iodination of nucleic acids using chloramine T was developed which allows for in vitro 125I-labeling of DNA and RNA. Labeled iodocytosine is shown to be the product of RNA modification. The labeled RNA preparations are non-degraded and possess high specific radioactivity. The iodination product is heat-stable and is insignificantly subjected to radiolysis.
