Preparation of spheroplasts from citrobacter freundii cells
The effect of various factors (age of the culture, pH, temperature, concentrations of EDTA, sucrose and lysosyme) on lysis of the Citrobacter freundii cells by lysozyme in the presence of EDTA was being studied. The spectra of the reaction mixture turbidity showed that the cells in Tris-HCl buffer are completely disrupted, whereas in the presence of 1.0 M sucrose formed mainly spheroplasts. A method, based on preparation of spheroplasts, was applied to localize tyrosine phenollyase in the cells of Citrobacter freundii.
