Production of freeze inhibitor polysaccharides of rye secale cereale cultivar rosen and barley hordeum vulgare cultivar hudson in tissue culture

Tissue cultures of Secale cereale L., ('Rosen' rye) and Hordeum vulgare ('Hudson' barley) were developed in order to produce larger quantities of freeze-inhibitor polysaccharides less expensively than can presently be done from cereal crown tissue. Culture techniques were perfected so that viable aseptic callus was obtained form rye and barley seed embryo with greater than 95% success. The callus was grown on a solid medium for approximately 4 weeks at 23.degree. C and then transferred to liquid shake culture, first at 23.degree. C and then at 0.degree. C. The freeze-inhibitor ratings of polysaccharides produced by rye and barley in tissue culture were similar to those of polysaccharides extracted from hardened crown tissues of the same cultivars. Arabinose and xylose constituted greater than 75% of the unpurified freeze-inhibitor polysaccharides produced in culture.
