Monolayers and multicellular spheroids of cultured Chinese hamster lung cancer V-79-4 cells were tested for their ability to survive ionizing radiation. Comparison of survival curves indicated a definite increase in radioresistance for the small spheroids. Using alkaline sucrose gradient sedimentation, DNA strand breakage repair was studied as a possible mechanism of survival. Single cells showed slower and less complete repair than cells in spheroids.
