Recent progress in the study on the mechanism of action of soybean lip oxygenase ec 1.13.11.12

Soybean lipoxygenase-1 (EC 1.13.11.12; MW 98,500) is a mononuclear non-heme Fe dioxygenase catalyzing the regio- and stereospecific oxygenation of polyunsaturated fatty acids containing a 1,4-cis,cis-pentadiene system. A review is given of some recent results. A new 1H NMR method to analyze the enantiomeric composition of fatty acid hydroperoxides is described. EPR studies of the NO complex of the enzyme indicate that the native enzyme contains Fe(II)-O2. Kinetic studies of the anaerobic reaction in which fatty acid hydroperoxides and fatty acids occur as substrates point to a substituted-enzyme mechanism with double substrate inhibition.
