Studies were designed to analyze the extent of strain- and/or species-specificity of suppressive factors of allergy (SFA) in selectively regulating Ig[immunoglobulin]E antibody production. The model system employed involved the previously described irradiation-enhanced IgE antibody response system of low and high IgE responder mice. SFA was obtained from complete Freund's adjuvant-induced ascites fluids from low or high IgE responder donor mice and then fractionated on concanavalin (Con)A Sepharose affinity columns. When fractionated this way, it was unambiguously demonstrated that SFA exerts selective effects on IgE antibody production across strain barriers in the mouse, irrespective of whether the recipient mouse is of low or high IgE responder type; more importantly, such Con A-Sepharose-fractionated SFA could be obtained from high IgE responder mice and could manifest their effects in suppressing IgE responses of high and low IgE responder recipients. Initiation of 2-way human mixed lymphocyte cultures (MLC) resulted in elaboration in such MLC supernatants of a biologic activity indistinguishable from murine SFA, whose biologic effects could be demonstrated in selectively suppressing IgE antibody production in irradiation-enhanced IgE responses of low responder mice. The implications of these observations in terms of providing relevant clinical alternatives for therapeutic amelioration of IgE-mediated diseases was discussed.
