Relationship between liver disturbance and mono amine oxidase a study of mono amine oxidase and isozyme of experimental liver disturbance

Liver disorders were induced in Wistar rats by the i.p. infusion of CCl4 i.p. and changes in monoamine oxidase (MAO) and histochemical characteristics of the liver were studied. MAO values in serum and soluble liver fractions increased in parallel with the liver hydroxyproline and with progression of liver fibrosis. The MAO of soluble fractions of the liver is apparently related to fiber formation. The serum MAO level increased comparatively early after CCl4 infusion, while the liver MAO level decreased. Although escape of liver MAO into the blood is indicated, MAO levels in the soluble fractions and mitochondrial fractions of the liver were high. Serum MAO of normal rats showed 9 bands in polyacrylamide gradient gel electrophoresis. Intensification of the 6th activity band was observed in serum MAO of rats with chronic liver disorders and this band coincided with the activity band of soluble fraction. The serum MAO which increases with fibrosis is apparently derived from the soluble fraction. Isozyme patterns of serum MAO of rats with acute liver disorders showed intensification of the 3rd activity band and the appearance of a new band between the 2nd and 3rd bands not observed in other groups. MAO may have originated in the liver mitochondria although it did not coincide with the isozyme of m-MAO.
