Reproducibility of nuclear measurements in computer assisted image analysis

Reproducibility in measuring nuclear features using microcomputer-assisted planimetry was evaluated. The area, perimeter, form factor and maximum diameter of 30 nuclei of cells of a poorly differentiated lymphocytic diffuse malignant lymphoma were measured using grey-scale level selection. Each of the eight observers performed the measurements twice. Intra-observer reproducibility was excellent for area, perimeter and maximum diameter, and was good for form factor results. Good inter-tobserver reproducibility for all parameters except form factor was also demonstrated. The high degree of variation among observers in measuring form factor may cause significant problems in the morphometric studies of tissues, especially when results from different laboratories are compared.
