Reproducibility of phase contrast microscope measurements of percentage motile microorganisms in samples removed from the dorsum of the tongue

The purpose of this investigation was to determine the accuracy of measurements of percentage motile microorganisms in samples removed from the dorsum of the tongue, using a phase-contrast microscope. The results indicated that the percentage of motile microorganisms in a bacterial sample can be accurately assessed provided that a short interval of time elapses between sampling and measuring. Furthermore, it appeared that the percentage of motile microorganisms varied during the course of the day.
