Serological detection of barley yellow mosaic virus wheat yellow mosaic virus and soil borne wheat mosaic virus by elisa enzyme linked immuno sorbent assay

Enzyme-linked immunsorbent assay (ELISA) was used to detect wheat yellow mosaic virus (WYMV), barley yellow mosaic virus (MYMV) and soil-borne wheat mosaic virus (SBWMV) in leaf and root extracts of infected barley and wheat. The .gamma.-globulin and enzyme conjugated .gamma.-globulin were used at 2 .mu.g/ml and 1:800 dilution, respectively. WYMV, BYMV and SBWMV were detected by ELISA in extracts of infected leaves diluted up to 25,600, 6400 and 12,800 times with phosphate buffered saline containing 0.05% Tween 20 (PBS-T), respectively. In the case of roots, WYMV and BYMV were detected in extracts with 0.1 M, pH 7.0 citrate buffer, but not with PBS-T. In ELISA using antisera against WYMV and BYMV, neither rice necrosis mosaic or oat mosaic viruses were detected. ELISA was developed to index a large number of plants of the breeding field in place of X-body or EM observation.
