Spectrophotometric determination of phenytoin in the presence of other anti convulsants

Phenytoin alone or in combination with either phenobarbital, diazepam, primidone or carbamazepine was added to normal human plasma processed for extraction and spectrophotometric determination of phenytoin. Phenobarbital, primidone and diazepam at therapeutic levels and above did not interfere in the estimation. A 2-3% interference on molar basis was observed with higher concentrations (20-50 .mu.g) of diazepam. Carbamazepine interfered at all concentrations tested giving about 11% contribution on molar basis. The oxidation products of phenytoin and carbamazepine gave similar spectra with a maxima at 247 nm. The product from diazepam had a maxima at 228 nm and residual absorbance at 247 nm.
