Conformational changes induced by changes of pH in guanyloribonuclease from Streptomyces aureofaciens RNase Sa were studied by circular dichroism. RNase Sa exhibited a complex negative circular dichroism around 280 nm with a clear fine structure and a positive Cotton effect at 235 nm. As circular dichroism around 280 nm is predominantly due to fixed tyrosine residues of RNase Sa, changes in their conformation and/or mobility were found to reflect clearly conformational changes of the enzyme. The protonization of tyrosines induced a negative Cotton effect at 240 nm, whereas after the ionization of tyrosines a positive band at 250 nm in circular dichroism spectra was observed. However, little conformational changes at pH 8.5 and weak conformational transition in the pH range from 5-3 were found. On the other hand, strong conformational changes at pH < 3 as well as pH > 10.5 were detected.
