Studies on cryptolepine 3. effect of cryptolepine on the tone and prostaglandin production in isolated rabbit duodenum

The inhibitory effects of cryptolepine [from Cryptolepis sanguinolenta] and its interaction with prostaglandin E2 [PGE2], indomethacin, adrenaline [epinephrine] and acetylcholine were investigated on the isolated rabbit duodenum. Cryptolepine (10-20 .mu.g/ml) or adrenaline (0.5-1 .mu.g/ml) or indomethacin (10-20 .mu.g/ml) abolished or reduced pendular contractions and relaxed the tissue. Cryptolepine (20 .mu.g/ml) produced an initial contraction before relaxation of the tissue. Cryptolepine (10-20 .mu.g/ml) abolished responses of the duodenum to added PGE2 and acetylcholine, but failed to reduce PG production. Cryptolepine apparently antagonizes PG, but has no inhibitory effect on its synthesis or release.
