Studies on genes and isoforms of myosin heavy chain from adult chicken skeletal muscle

Total RNA was isolated from adult chicken breast muscle, and RNA containing poly (A) was purified from total RNA by chromatography on oligo (dT)-cellulose. 28S-32S poly (A) RNA containing myosin heavy chain mRNA was purified from poly (A) RNA by the sucrose density-gradient centrifugation method. Double stranded cDNAs were constructed using this 28S-32S poly (A) RNA. Poly (dC) tailed cDNA was hybridized with poly (dG) tailed pBR322 and used to transform the Escherichia coli K-12 strain .chi.1776, and the cDNA library of adult chicken breast muscle was prepared. A group of synthetic oligonucleotides (17-mer and 18-mer), based on the amino acid sequences of the portions of 25KDa, 50KDa, 20KDa of myosin heavy chain respectively, was constructed. The library was screened by colony hybridization with a group of synthetic oligonucleotides 32P-labeled as a probe. The resulting two cDNA clones, clone 1-20-18 and clone 1-20-44, cover the S-1 region. The deduced amino acid sequence of clone 1-20-18 agreed with the amino acid sequence of the protein expressed as myosin heavy chain in the breast muscle. On the other hand, other total RNA was isolated from adult chicken leg muscle. In the same way, the cDNA library was prepared. This library was screened with clone 65 constructed previously, the cDNA clone of myosin heavy chain in embryo chicken leg muscle, as a probe. The resulting four cDNA clones, based on the carboxyl-terminal sequence for LMM region of myosin heavy chain, were selected. On the result, it is suggested that at least two isoforms of myosin heavy chain mRNAs exist in adult chicken breast muscle, and at least four isoforms of myosin heavy chain mRNAs exist in adult chicken leg muscle.
