From postheparin plasma, hepatic triglyceride lipase (H-TGL) was completely separated from lipoprotein lipase (LPL) by means of affinity chromatography on heparin-coupled Sepharose. The antiserum against this partially purified H-TGL was produced in a rabbit. By using this antiserum, H-TGL but not LPL, was completely inactivated, providing a new method of selective determination of H-TGL and LPL activities in human postheparin plasma. The method described here was more specific and reproducible when compared to the conventional method using 1 M NaCl or protamine. Using the immunochemical method, H-TGL and LPL activities were measured in the postheparin plasma obtained from 15 normal subjects. The present immunochemical method is useful in studies on abnormalities of the metabolism of various lipids in man.
