Study of factors affecting the structural and functional properties of photosystem ii particles with an active water splitting system from the membranes of the thermophilic cyanobacteria synechococcus elongatus

A comparative study of the O2-evolving activity, the content of P700 admixtures, and low-temperature absorbtion and fluorescence spectra has been carried out for the photosystem (PS) II particles from the Synechococcus elongatus cells in the process of seasonal changes in the culture growth. The decrease in the electron transport rate in the isolated PS II particles concomitant with the decelleration of the cell growth was found to be caused by the decline in the photosynthetic apparatus stability. Isolation of particles in the presence of 25% glycerol made possible a considerable restoration of their functional activity. The results suggest that preservation and efficacy of the functional coupling with the reaction centers of light harvesting phycobillin complexes determine the heterogeneity of the preparations obtained upon isolation and incubation of the PS particles, in particular their size and different rates of the electron transport.
