The blocking of phosphodiester (PDE) groups of oligonucleotides was investigated by pulsed 31P-NMR spectroscopy. Interaction of (Tr)TpT(Ac) with.beta.-cyanoethanol or aniline in the presence of TPS or the triphenyl phosphine-CCl4 pair, respectively, proceeds via the intermediate formation of P1,P2-bis-5'-O-tritylthymidine-bis-3'-O-acetylthymidine-5'-pyrophosphate which further converts into (Tr)TpT(Ac).beta.-cyanoethyl ester or anilide.
