The effect of adenine and mevalonic acid on the endogenous cyto kinins of a cyto kinin dependent strain of soybean glycine max cultivar acme callus

The combined application of 10-6 M adenine and 10-6 M mevalonic acid to soybean callus accelerated its growth. Two biologically active compounds that co-chromatographed with zeatin and isopentenyl adenine were extracted from this callus. Studies with labeled adenine and mevalonic acid indicated that the cytokinin-dependent soybean callus incorporated only a very small amount of the radioactive precursors into the biologically-active compounds, making it extremely difficult to determine whether these compounds were synthesized de novo or whether they arose as by-products of tRNA turnover. As cytokinins do not accumulate in rapidly-growing cytokinin-dependent soybean callus cultured on kinetin as a source of cytokinin, it seems as if biosynthesis de novo occurs when the callus is supplied with adenine and mevalonic acid.
