The formation of a root specific iso peroxidase as an indicator of root primordium differentiation in pith of tobacco nicotiana tabacum cultivar samsun

The isoperoxidases in excised pith of tobacco plant (N. tabacum cv. 'Samsun') was examined by starch-gel electrophoresis. In cultured pith pieces, 4 cathodic isoperoxidases designated Cx, Cy, C4 and C5 developed within 2 days after inoculation. In 6 cm long stem section taken from the internode at 12-24 cm of distance from apex, a root-specific isoperoxidase C5 was induced only in the original basal pole, while the 3 others were induced in both apical and basal poles. The difference may demonstrate C5 to be induced in response not to injury but to the change which would reflect a certain physiological change leading to differentiation of root primodium. Actinomycin D did not inhibit the development of injury-dependent isoperoxidases but completely inhibited the development of C5. In the presence of 2, 4-D in the medium, C5 was never developed in excised pith without significant diminition of ininjury-dependent isoperoxidases.
