A search was made for strand-breaks in the DNA of nuclei in different regions of the root cap of Z. mays using terminal deoxynucleotidyl transferase and 3H-dCTP. Quantitative autoradiography of the resulting reaction product showed that DNA strand-breaks do not accumulate as the cells pass from a proliferative state in the cap meristem to a degenerative state on the outer surface of the cap. A microdensitometric investigation of root cap nuclei with a 4 C DNA content showed that the proportion of dense chromatin is higher in the outer cap cells than it is in either the central or meristematic cells.
