The primary structure of snake zaocys dhumnades dhumnades insulin
Snake insulin was isolated and crystallized from the pancreas of a non-venomous snake (Z. dhumnades dhumnades). The crystalline form is not rhombohedral but dodecahedral. The primary structure was determined with the aid of an LKB solid-phase sequencer. In the primary structure of snake insulin, the presence of B5Arg, B29Arg, B16Phe, B25Tyr and B18Ile is unusual in comparison with insulins from mammals, birds and fishes. The snake insulin studied is very similar to rattle snake insulin, with the exception that A15 is Glu instead of Glu and B30 is Thr instead of Ser. These differences are consistent with the higher acidity of the snake insulin studied and with the Thr and Ser content in amino acid analysis. The B10 residue of the snake insulin is still histidine. The formation of dodecahedral crystals of the snake insulin was discussed in connection with the presence of this histidine.
