The subcellular localization of a glucuronyltransferase involved in glucuronoxylan biosynthesis in pea pisum sativum epicotyls

The subcellular location of a glucuronyltransferase (GT) involved in glucuronoxylan biosynthesis in pea (Pisum sativum) has been investigated using continuous and discontinuous sucrose-density gradient centrifugation. The densities of membranes from different organelles were determined by locating on the gradients their specific marker enzymes, namely NADH-cytochrome c-reductase for endoplasmic reticulum, Triton X-100-activated IDPase and Glucan synthetase I for Golgi apparatus and Glucan synthetase II for plasma membrane. GT activity (both in the presence and absence of UDP-D-xylose) was found to be located in the Golgi apparatus.
