Tumor inhibition by metallocenes anti tumor activity of niobocene di chloride and tungstocene di chloride

The anticancer activities of (C5H5)2NbCl2 (NDC) and (C5H5)2WCl2 (WDC) are investigated against Ehrlich ascites tumor cells in CF1 mice. With the W(IV) metallocene WDC only sporadic tumor inhibition is observed; the paramagnetic Nb(IV) derivative NDC is a potent cancerostatic agent causing 100% cures in doses of 20 and 25 mg/kg. The side effects of NDC (LD50 = 35 mg/kg) are considerable and unique in the series of cancerostatic metallocene dihalides. Air oxidation of NDC yields the diamagnetic binuclear Nb(V) complex [(C5H5)2NbCl2]2O which has reduced toxic, but also diminished tumor-inhibiting, properties.
