Ultrastructural changes in myocardium induced by a single dose of doxorubicin in rabbits
The effects of a single bolus injection of doxorubicin (DXR) on myocardial cell fine structures were studied on male albino rabbits. (Group A: 20 mg/kg) Typical nucleolar segregation had already appeared 1 hr after the injection, and after 3 hr, such findings were more prominently observed. After 3 and 10 hr, contraction band formation and dissociation of intercalated discs were seen, and after 24 and 48 hr, separation of myofibrils and augmentation of lipid droplets were observed. (Group B: 10 mg/kg) Although 3 hr after the injection the nucleolus showed typical or partial nucleolar segregation, the nucleolus appeared to be normal in structure 4 d later. After 4 and 7 d, marked augmentation of lipid droplets, separation of myofibrils, mitochondrial swelling and myelin figure formation were observed. (Group C: 5 mg/kg) Three hr after the injection, the nucleolus showed partial nucleolar segregation. Lysis of myofilaments and dilatation of the sarcoplasmic reticulum were observed 3 w after, and progressed over a period of time, while a pronounced myofibrillar disorganization and the "adria" cell formation were seen 8 w after the injection. The impairment of nuclear DNA by DXR may play a significant role in the development of DXR-induced cardiomyopathy. On the other hand, various types of myocardial changes observed in the early stage of the experiment may be induced by other complex mechanisms of the drug.
