Ultrastructural changes in smooth ended and ruffle ended maturing ameloblasts of rats after vinblastine administration

Young male Wistar rats were given intravenous administrations of 2 mg/kg body weight of vinblastine sulfate (VBL). After the passage of 3, 6, 24, and 48 hr cytological changes in smooth-ended and ruffle-ended ameloblasts (SAs and RAs) were observed. Characteristic changes occurring 3 hr after VBL administration included reduction in numbers of microtubules, disturbed cell polarity caused by movement of organelles and nuclei, and shortening of microvillous projections in the RA ruffled border. Unusual accumulation of secretion granules and disruption of the rough-ER occurred 6 hr after administration. After 24 hr, deteriorative changes occurred in the mitochondria. But these kinds of cellular damage had all recovered 48 hr after administration. The SAs were more sensitive to VBL than the RAs and seemed to have disappeared as a result of vacuolization 6 hr after administration. SAs observed 24 hr after administration were presumed originally having been RAs that were morphologically modified during the recovery process.
