The role of Ca2+ ions in regulation of Na+,K+-ATPase activity by endogenous regulators has been studied in saponin-permeabilized red blood cells of rat. It was shown that using oubain for determination of Na+,K+-ATPase activity in the presence of Ca2+ leads to errors resulting from the Ca2+-dependent action of oubain on Ca2+-ATPase. Oubain (1 mM) activates Ca2+-ATPase at low Ca2+ concentrations (0.2-0.7 .mu.M) and inhibits it at higher Ca2+ concentrations. In the Na+-depleted media with Ca2+-Mg2+-EGTA-ATP-citrate buffers, at 50 .mu.M Mg2+, inhibition of Na+,K+-ATPase by Ca2+ reached 100% at 1-2 .mu.M Ca2+. In the presence of 370 .mu.M Mg2+, corresponding to its intracellular concentration, 0.2-0.7 .mu.M Ca2+ activated Na+,K+-ATPase, while at Ca2+ concentrations above 1-2 .mu.M a partial inhibition of Na+,K+-ATPase was observed. Washing the membranes with EGTA-containing media eliminated both effects of Ca2+. The participation of Ca2+-dependent proteins and protein kinases in regulation of Na+,K+-ATPase activity is discussed.
