Asymmetric somatic hybrid calli between leguminous species of lotus corniculatus and glycine max and regenerated plants from the calli

Protoplast fusion was performed between birdsfoot trefoil (Lotus corniculatus L.) and soybean (Glycine max (L.) MERR.). In order to produce asymmetric hybrids containing a complete birdsfoot trefoil genome and a small part of a soybean genome, or cybrids containing only a birdsfoot trefoil genome, iodoacetamide-treated protoplasts of birdsfoot trefoil were fused with X-ray-irradiated soybean protoplasts. Peroxidase isozyme and karyotypes of calli obtained from the protoplast fusion were analyzed and as a result, the hybridity of some of the calli was elucidated. Plant regeneration from the asymmetric hybrid calli was also successful, but regenerated plants did not show the hybridity as a result of analysis of peroxidase isozyme. This result may be caused by the elimination of soybean chromosomes. The morphology of the regenerated plants resembled that of birdsfoot trefoil derived from parent calli. However, the regenerated plants were usually teratologically an erect type in contrast to the creeping type of normal birdsfoot trefoil.
