Owing to lack of African green monkey (Cercopithecus Aethiopos) and lower passage cell line BSC-1 or CV-1, SV40 was failed to be detected. Thus polio vaccine production in our country was not in conformity to WHO requirements of polio vaccine quality. A large number of rhesus monkey used for the production was discarded due to SV40 contamination in past years. This paper reports a modified counter immunoelectrophoresis for detection of SV40. The method is rapid, sensitive and specific for SV40 detection without need of any cell line. Altogether 70 SV40 samples verified by immunoelectromicroscope (IEM) and neutralization test (NT) were found to be positive by this method without any false positive. The method has been applied the production of polio vaccine in our institute in 1985 with success.
