Cyclodextrin glucanotransferase (.alpha.-1,4-glucan 4-glycosyl transferase, EC 2.4.1.19; CGTase) is an unique enzyme wiht the ability to convert starch to cyclodextrin (homogeneous cyclic molecules of six or more .alpha.-D-glucopyranose units linked 1-4 as in amylose). An ultraviolet induced mutant, No.35, capable of producing large amount of CGTase in an alkaline medium has been obtained from an alkalophilic Bacillus spl. The CGTase from the mutant was able to convert starch to beta and gamma cyclodextrins at the ratio of 9 to 1. However, no alpha-form was detected. A medium containing 3.5% starch, 0.5% urea, 1.0% hydrolyzed plant protein, 0.5% rice bran and 1.0% sodium carbonate was found suitable for the enzyme production. The maximum activity, 2.199 units/ml, could be obtained 24 hours after inoculation in a 5 1 jar fermentation with pH value controlled at 8.0. When 10% crude CGTase was added into 10% and 15% starch solution, the yields of beta-cyclodextrin reached 68% and 38%, respectively, after 48 hr incubation.
