Dauricine (Dau), an effective antiarrhythmic drug, produced concentration-dependent depression on the APA, .ovrhdot.Vmax and SR in human atrial fibers and rabbit atrioventricular node cells and on SP4 only in human atrial fibers, and prolonged APD90 only in rabbit atrioventricular node cells. The effects of lidocaine (Lid) on all parameters of action potential were insignificant at the treatment concentration. During the perfusion with 100 .mu.mol/L Lid, the .ovrhdot.Vmax and APA were decreased in human atrial fibers, and with 30 .mu.mol/L, .ovrhdot.Vmax and SP4 were reduced in rabbit atrioventricular node cells. In both preparations perfused with Lid, SR tended to decrease. Dau markedly increased SACT of the anesthetized rabbits, but had no significant effects on SNRT, CSNRT, SNRTI and HR. The action of Dau, when administered in combination with Lid, on SACT was not greater than that produced by Dau alone, but CSNRT was significantly increased. The results suggest that Dau significantly depressed the sinoatrial node conduction function, and the action potential of the atrial fibers and atrioventricular node cells, which might be related to its antagonizing effect on supraventricular arrhythmias.
