The distribution of alkaline- and acid phosphatase activity in different stages of oocyte growth and differentiation in Puntius sarana has been studied. Alkaline phosphatase activity is moderate in the nucleus and nucleoli of early oocytes, but weak in the cytoplasm. The enzyme activity in the cytoplasm gradually increases in the subsequent growth stages and attains its peak in the late maturing oocytes and thereafter there is a decrease. Intense activity of alkaline phosphatase in the cortical cytoplasm and follicular epithelium of maturing (vitellogenic) oocytes suggests its role in the transport of yolk precursors across the oocyte membrane and in the synthesis of proteins and/or steroid hormones. The resrobing oocytes show acid phosphatase activity in their yolk bodies. The activity of the enzyme is intense in the granulosa cells during their phagocytic activity. These observations indicate that the lysosomal activity facilitates the easy resorption of yolk. The normal, healthy oocytes (immature to mature stages), however, reveal little enzyme activity.
