The possibility of using antisense RNA to inhibit the expression of neomycin phosphotransferase II [NPT] gene in tobacco protoplasts was verified. The construction of the recombinant plasmids pB4 and pVIL35 was described. NPT activity was determined, using gel electrophoresis and radioautography. The results of the study confirm the possibility of using antisense RNAs for highly effective inhibition of gene expression. Plants resistant to various diseases can be created by incorporation of various recombinant structures with antisense RNAs of pathogenicity genes.
