Isolation and determination of cell membrane protein antigen from cultured lymphoid tumor cells from calf type of bovine leukosis

In the cultured tumor cell line, BTL-C 3, from calf type of bovine leukosis, bovine T-lymphoid tumor cell surface antigen (BTLA) was detected strongly. In the present experiment, soluble protein antigen from the cell membrane of BTL-C 3 cell line was studied. The tumor cells were homogenized in teflon tube, and cell membrane was isolated by sucrose density gradient centrifugation (100,000 g, 60 min). The cell membrane was isolated around density = 1.17 fraction and treated with NP 40 to be soluble. The soluble protein antigens were separated by SDS PAGE-Western blotting method, and were detected as 30 to 40 bands. Three bands with molecular weights of 50,000, 35,000 and 27,000 respectively, showed specific reaction with the anti-BTLA serum. These bands were considered to be major components of BTLA.
