One Marek's disease virus (MDV) field isolate, designated Z4, was identified as serotype II MDV. On primary isolation, Z4 virus grew slowly in chicken embryo fibroblast (CEF) cell cultures. Typical plaque characterized by rounded refractile degenerating cell and some large syncytia did not develop until the 12th day post-inoculation. Susceptible day-old chicks intraabdominally inoculated with 1000 pfu Z4 virus did not develop signs and lesions of MD, although mild inflammatory infiltration in peripheral nerves of some inoculated chicks were occasionally found. Birds inoculated at 1-day-old with Z4 virus were resistant to the oncogenic effect of subsequent challenge with virulent MDV GA strain at 1-week old. These promising features suggested that Z4 should be a good candidate of vaccine virus against MD. In addition to Z4, other 11 MDV isolates obtained in China, including a virulent virus isolated in Beijing and an apathogenic virus isolated in Harbin were also serotyped with type specific monoclonal antibodies. Of these 11 isolates, 7 were serotype I, 2 were serotype III and the other 2 isolates contained both serotype I and serotype III viruses. The results offered preliminary information the prevelence and distribution of different serotypes of MDV in chicken flocks in China.
