Light microscopic morphology and ultrastructure of the proximal tibial growth plate in experimental diabetic rats

The proximal tibial growth plate in streptozotocin-induced diabetic rats (n=20) was studied histopathologically. The thickness of the proximal growth plate was significantly less in diabetic rats than in the controls (n=25) by the end of the 16th week after induction. Chondrocyte columns were irregular and the chondrocytes themselves small and irregular in shape in diabetic animals, and the pericellular matrix revealed lower concentrations of proteoglycan in the matrix. Electron microscopic observations showed severe degenerative changes in chondrocytes located in the hypertrophic zone. These included numerous microfilaments, large lipid bodies and poorly developed rough endoplasmic reticulum. Longitudinal bone growth was significantly reduced in the diabetic group (p < 0.01). These findings suggest that impaired endochondral ossification associated with degeneration of hypertrophic cells occurs in the induced diabetic state in rats.
