Nuclear behavior during basidiospore formation and germination of the ectomycorrhizal fungus tricholoma robustum

Nuclear behavior of the ectomycorrhizal fungus Tricholoma robustum during basidiospore formation and germination was examined by fluorescence microscopy. During basidiospore formation, two nuclei first fused within the basidium followed by meiosis. Subsequently, each of the four nuclei migrated into one of the four developing basidiospores. Within basidiospores, the third nuclear division (post-meiotic mitosis) occurred, and one of the daughter nuclei returned into the basidium. Consequently, mature basidiospores each contained a single nucleus. During germination two patterns of nuclear behavior were observed: (A), the first nuclear division occurred within basidiospores, and one of the daughter nuclei migrated into the germ tube; (B), The nucleus migrated into germ tube followed by the first nuclear division. Pattern A was more frequently observed than pattern B. The monokaryotic state was steadily maintained from the initial stage of germination.
