Pharmacological modulation of endogenous norepinephrine release from cardiac sympathetic nerves

The sympathetic regulation of neurotransmitter release was investigated on isolated rat heart with the supply of the right sympathetic chain. The electrical stimulation(ES) of the post-gangionic cardioaccelerator nerve produced an increase of the rate of contraction, likely related to the ES rate. Norepinephrine (NE) release was measured by collecting the coronary perfusate and by a direct analysis on an HPLC-Electrochemical detector apparatus. The evoked NE outflow was increased by drugs acting at pre-synaptic level like desipramine, which is a neuronal re-uptake inhibitor, or yohimbine which antagonizes the alpha-2 adrenergic recepter. Both drugs did not modify the induced tachycardia. Beta-adrenergic blockade with propronaolol completely abolished the increase of the contraction rate due to stimulation, despite the presence of high levels of NE in the coronary perfusate. This "organ-nerve" preparation appears useful for studying the activity of sympathetic nervous system in regulating myocardial function and neurotransmitter release.
