Polyacrylamide gradient gel electrophoresis a routine method for high resolution isoenzyme electrophoresis of solanum and lycopersicon species

This paper describes a refined method for isozyme analysis based upon polyacrylamide gel electrophoresis. The method could be routinely used and clearly reproducible banding patterns were obtained for Pgdh, Pgm, Pgi and Got in Lycopersicon peruvianum, Solanum lycopersicoides, Solanum pennellii and tuber-bearing Solanum species hybrids. When compared to starch gel electrophoresis, the present method is superior in giving more constant and high quality bands with a better resolution of isozyme variants. In addition, pore size and pH gradients may be applied within a single gel to optimize band separation and resolution. This is of particular importance when there are only subtle differences in the migrational capacities of different isozyme variants which cannot be otherwise resolved.
